In an interview with Metro (via Shocktillyoudrop), Aykroyd says Bill Murray is not going to be involved with Ghostbusters 3.

“It's sad but we're passing it on to a new generation. Ghostbusters 3 can be a successful movie without Bill. My preference would be to have him involved but at this point he doesn't seem to be coming and we have to move on. It's time to make the third one,” Aykroyd told Metro.

As far as I know, the movie can only be made with Aykroyd, Murray, Harold Ramis, and Ivan Reitman all agreeing to do so since each of them have final script and movie approval. Murray was never fond of the second movie in the first place, and he's even less enthused about a third one.

For whatever reason, Murray, the lone holdup, has finally given in and allow the movie to proceed without him. If that's the case, Ghosterbusters 3 is definitely happening.

As Aykroyd said above, the third movie will be about the Ghostbusters passing their proton packs to a new generation. If the movie's successful, we'll see Ghostbusters 4 and beyond with these new characters. This is really what Ghostbusters 3 comes down to – to keep the franchise going.

The latest draft of the script is being worked on by Men in Black 3's Etan Cohen.
